| Updates - Family Medical History | Update received August 2025: Donor's Father underwent atrial fibrillation cauterization at age 60, he was also diagnosed with high blood pressure and high cholesterol at age 60, treated with medication; Donor's Sister was diagnosed with uterine fibroids and ovarian cysts at age 30, treatment unreported; Maternal Grandmother was diagnosed with high blood pressure and high cholesterol at age 80 and macular degeneration at age 83, treatment unreported for all; Maternal Uncle #1 has died, age and cause of death unreported; Paternal Grandmother has died at age 70 from a C. diff infection.UPDATE Sept 2025: Donor's Daughter #1 was diagnosed with anemia and Type 1 Von Willebrand disease (VWD) at age 13, treatment unreported. Follow up with the donor lead to the following updates - his youngest daughter also tested positive for vWD through factor testing (not genetics) but has not experienced any symptoms while his oldest daughter takes Humate-P infusions weekly to support her clotting and prevent pain in her joints from microbleeds. Of note, this donor has 22 donor conceivedchildren with no reported medical issues.UPDATE October 2025: The donor, though asked, did not confirm if he had bloodwork to determine if the inheritance of his children’s vWD was autosomal dominant or recessive nor if any extended family history of a similar diagnosis exists.vWD1 is typically caused by a single variant in the VWF gene and is inherited in an autosomal dominant manner. Many people with autosomal dominant vWD have an affected parent. Based on the reported family history, it is likely the vWD1 cases reported follow autosomaldominant inheritance, however it is unclear if the donor or his wife has the condition. The proportion of individuals with type 1 VWD who have a VWD-causing variant identified is lower than other forms of VWD. As genetic test results were not available on the affected child, a negative genetic test on the donor would not be informative. |
