He carries a mild (S) variant and children are only at risk of adult-onset disease if the egg source is a carrier for a severe (Z) variant in the SERPINA1 gene.
Updates - Family Medical HistoryUPDATE September 2025: A pregnancy was identified to have a genetic variant in the FLG gene consistent with a diagnosis of ichthyosis vulgaris. The egg source tested negative. It is unknown if this variant was inherited from the donor or is new to this pregnancy. Donor testing is pending. This donor has met his family unit limit - no other children conceived with the help of this donor have reported a medical update or diagnosis. An update will be made available once the donor's results are complete.UPDATE September 2025: The donor was subsequently identified to carry this variant. All offspring conceived by this donor have a 50% chance to inherit this variant. Ichthyosis vulgaris is a skin condition that leads to dry, scaly skin which is usually most severe on the legs but can also involve other parts of the body including the ears, which can lead to hearing loss. The skin changes in ichthyosis vulgaris tend to appear in the first year of life though rarely affect a person’s overall health. The condition usually improves during adulthood but may return as people age. No offspring have been reported to show signs of this condition.
