Updates - Family Medical HistoryUPDATE July 2025: A pregnancy conceived using donor 6357 was diagnosed with a ventricular septal defect and spina bifida with associated intracranial findings include ventriculomegaly, strawberry shaped skull, banana shaped cerebellum and clubfoot at 19 weeks gestation. Per the client’s medical records, prenatal cell-free DNA screening was low risk for trisomy 21, 18, 13, sex chromosome aneuploidy, triploidy, and 22q11.2 deletion. Amniocentesis with whole exome sequencing and chromosome microarray analysis was negative. These results reduce the chance of an underlying genetic etiology, however that risk cannot be eliminated. Without knowing the underlying cause for these medical issues, we are unable to provide a risk for recurrence. A detailed anatomy scan and a fetal echocardiogram are recommended during any pregnancy conceived with the help of this donor. Update received September 2025: Maternal Grandfather has died from pulmonary failure at age 88.UPDATE Dec 2025: Fairfax was notified that a client conceived fraternal twins. One twin was noted to have ventriculomegaly on prenatal ultrasound at approximately 20 weeks’ gestation, which reportedly resolved postnatally by around 4 months of age.This same child was later diagnosed with bilateral cataracts at approximately 1 year of age and underwent surgical correction. Per the client, infectious studies (a known cause of both ventriculomegaly and cataracts) were negative. The child is reported to have developmental delay; however, the client is uncertain whether this is attributable to a period of impaired vision. Genetic testing to potentially identify a hereditary cause of these findings has not been conducted on the child. Without further clinical evaluation or investigation of an underlying etiology, inheritance and a specific risk for recurrence in other donor-conceived children remains unknown. A confirmed genetic contribution has not been associated withwith this donor. This donor has met his family unit limit.
