Update received July 2024: Paternal Grandmother has died at age 88 from an accidental fall.Update January 2025: A child was born with a unilateral cleft lip and a double aortic arch. The co-occurrence of congenital heart disease and cleft lip, with or without cleft palate has been reported in multiple studies, suggesting that patients with orofacial clefting have an increased risk of congenital heart disease with risks ranging from 8-30% in these reports. The child had extensive genetic testing completed including a chromosomal microarray and exome array which failed to identify any chromosome changes or gene variants associated with a genetic syndrome consistent with these physical features. The risk for other half siblings to have a cleft lip and/or a CHD present at birth is slightly higher than the general population risk and is believed to be about 3%. A detailed anatomy scan and a fetal echocardiogram are recommended during any pregnancy conceived with the help of this donor. Update May 2025: A child was reported to have a low-grade glioma in the brain. As the child was not identified to have a pathogenic genetic variant, including any in the NF1 gene, the cause remains unknown. Genetic testing identified a variant of unknown significance in the ALK gene for which the mother tested negative. When a VUS is identified, it is unclear whether that variant is associated with a specific health condition. In many cases, the variant is so rare in the general population that it has not been described in the literature or variant databases,thus there is little published information to review. Identification of a heterozygous variant of uncertain significance does not establish or rule out the diagnosis of a disorder and no medical management recommendations are made based solely on finding of a VUS. As such, the donor was not tested for this VUS as it would not impact the clinical management of this case or his eligibility as a donor.Update July 2025: A recipient reported that her child was born with a double aortic arch. The child otherwise healthy. No genetic evaluation or testing has occurred at this point. Risk for recurrence in other donor-conceived people is believed to be increased. Without a know etiology, a specific risk of recurrence cannot be provided. A detailed anatomy scan and a fetal echocardiogram are recommended during any pregnancy conceived with the help of this donor. Update received September 2025: Maternal Grandfather has died at age 92, specific cause of death unreported; Paternal Grandfather has died at age 89 from lung cancer; Paternal Grandmother was diagnosed with dementia at age 87 prior to her death at age 88. |
