Updates - Family Medical HistoryNo change to donor Family Medical History: August 2023, September 2025.Update May 2025 - A pregnancy conceived with the help of this donor was identified to have an increased nuchal translucency, bilateral neck cysts, and generalizededema. The anatomy ultrasound and fetal echocardiogram was reportedly normal. Genetic testing identified a variant of uncertain significance (VUS) in the SOS2 gene,for which the donor was confirmed to carry.  When a VUS is identified, it is unclear whether that variant is associated with a specific health condition. In many cases, thevariant is so rare in the general population that it has not been described in the literature or variant databases, thus there is little published information to review.Identification of a VUS does not establish or rule out a diagnosis of a condition and no medical management recommendations are made based solely on the finding of aVUS. Given the current classification of this variant, the number of births reported for this donor, and the lack of additional medical reports for children conceived by thisdonor, our Medical Director and pediatric geneticist is allowing him to continue in our program. Update August 2025 - A client reports that her son presented with the following prenatal findings: increased nuchal fold (measured at 8.1 mm), a ventricular septal defect (VSD), which spontaneously resolved by later echocardiograms, and an echogenic intracardiac focus (EIF) in the left ventricle. Per the client, additional tests including karyotyping, SNP microarray, and a targeted Noonansyndrome panel (which included a 19 gene panel covering SOS2 among other commonly associatedgenes) were normal. Results were not available to Fairfax Cryobank at the time of this report. Per the client, the child is currently developing normally with no diagnosed conditions.UPDATE August 2025: Regarding the pregnancy identified to have the SOS2 VUS, the client reported that her baby was born eariler this year with no diagnoses and that they are currently meeting all milestones.
