Any child conceived with the help of this donor has a 50% chance to inherit this VUS. The potential impact of having this VUS remains unknown.
Updates - Family Medical HistoryUPDATE February 2026: Child born with possible POLRMT-related condition. Donor placed in Investigation while additional information is obtained. UPDATE March 2026: The health issues reported in the donor-conceived child include issues identified shortly after birth, such as difficulty maintaining normal body chemistry, breathing problems requiring medical support, and being smaller than expected. Doctors also noted differences in the baby’s muscle tone and reflexes, feeding challenges, abnormal heart valve function, and abnormalities seen on brain imaging and EEG testing. Genetic testing on the child identified a genetic change, called a variant of unknown significance (VUS), in a gene known as POLRMT. A VUS means that a genetic change was found, but it is not currently known whether the change is related to a medical condition. In many cases, these variants are very rare and have not been studied enough to determine their impact. The presence of a VUS does not confirm or rule out a medical diagnosis. At this time, no clear link has been established between this VUS and the child’s medical condition. Typically, for a child to be affected with a condition related to this gene, the child would need to inherit a variant from both the egg and sperm source to be affected, although there are situations where a single variant may be associated with health concerns. Prior to the donor’s passing, he provided a DNA sample to determine whether he carried the same variant identified in the child. These test results are currently pending. Of note, five other births have been reported from this donor with no medical concerns.UPDATE March 2026: Donor has been confirmed to carry variant of uncertain significance in the POLRMT gene that was identified in a donor-conceived child.
