UPDATE June 2026: Fairfax was notified that a child conceived with the help of this donor was identified to have a genetic variant associated with neurofibromatosis and that the egg source was not identified to have this variant. Genetic test results for the child and the egg source were requested. The donor was placed in Investigation while we obtain additional information regarding this medical update. UPDATE June 2026: The client mentioned above shared the genetic test results from her child and herself, confirming the presence of a pathogenic variant in the NF1 gene in the child that was not identified in the egg source. Approximately half of affected individuals have NF1 as the result of a new NF1 disease-causing variant that was not inherited from either the egg or sperm source. Fairfax will move towards consenting and testing the donor for the variant identified in this child. No timeline for this process is currently available. UPDATE June 2026: Variant specific testing for NF1 was ordered on the donor. Results are expected in approximately one month. UPDATE July 2026: A semen sample was sent to the testing laboratory for DNA extraction and targeted variant testing and was negative, i.e. the variant previously identified in the child mentioned above was NOT identified in DNA extracted from the donor’s sperm. For a couple with one child with an apparently de novo (new) NF1 pathogenic variant, if neither parent has clinical signs of NF1 and neither parent has the variant detected in blood, the recurrence risk for a future pregnancy is generally quoted as less than 1% because of the possibility of gonadal mosaicism. Gonadal mosaicism is a rare genetic phenomenon where a pathogenic variant is present in only a subset of the reproductive cells (egg or sperm cells), but not in other body cells. As testing was conducted on DNA extracted from the donor’s sperm, the risk is believed to be at least equivocal, if not less than this.
