| Updates - Medical | UPDATE Jan 2026: Donor 7659 contacted Fairfax to inform us that he was identified to carry a pathogenic variant (EX11del) in the PMS2 gene. This gene is most often associated with Lynch syndrome, which is a hereditary cancer syndrome. Per the donor, he and his mother were tested after his maternal uncle was diagnosed with colon cancer. The donor denies any additional family history of cancer. In rare cases, changes in the PMS2 gene can also cause a different condition called constitutional mismatch repair deficiency (CMMRD). PMS2-related Lynch syndrome follows autosomal dominant inheritance. There is a 50% chance that any child conceived with the help of this donor will inherit his PMS2 variant; those that do have an increased risk for colon cancer (8.7-20% lifetime risk) and endometrial cancer (13-26% lifetime risk in females). Individuals of reproductive age are at 25% risk of having a child with CMMRD with each pregnancy when both the sperm and egg source have a pathogenic variant in PMS2. |
